Function: It is a retro-inverso peptide designed to interfere with the p53-FOXO4 interaction, allowing p53 to drive apoptosis specifically in senescent cells.
Mechanism: Senescent cells often use FOXO4 to prevent p53 from inducing death, which helps them survive; FOXO4-DRI disrupts this, forcing the cells to die.

FOXO4-DRI (or Proxofim) is an experimental, synthetic senolytic peptide that induces apoptosis (programmed cell death) in senescent “zombie” cells.

By blocking the interaction between FOXO4 and p53, it causes p53 to trigger cell death, potentially reducing age-related damage and improving tissue function.

Studies indicate it can selectively target damaged cells.
